Which standard covers pressure differential testing?

ISO 14644-3 includes the installed pressure difference test among its cleanroom test methods. Pharmaceutical acceptance criteria normally come from EU GMP Annex 1, WHO guidance on HVAC systems and Schedule M, together with the facility's HVAC design. The approved protocol should state the required differential and direction for each pair of rooms.

What is the difference between a positive and a negative cascade?

In a positive cascade the cleanest room has the highest pressure, so air flows outwards and keeps contamination out. In a negative or sink arrangement, a room or airlock is kept at lower pressure to stop potent, toxic or biological material escaping. Many facilities combine both, using airlocks as pressure bubbles or sinks.

Why do room pressure gauges need to be verified?

Installed differential pressure gauges drift with time and can be affected by blocked or disconnected tubing. If they are wrong, daily monitoring records give false assurance. During validation each gauge is compared with a calibrated reference manometer at the same points, and any gauge outside tolerance is flagged for calibration or replacement.

How is airflow direction confirmed between rooms?

Besides the pressure reading, airflow direction can be shown with a smoke stick or fog source at a slightly opened door or transfer hatch. This gives visual evidence that air moves from the higher-pressure room into the lower one, and is useful where differentials are small or close to the acceptance limit.
