How are air changes per hour calculated in a cleanroom?

The total supply air volume entering a room is measured at each terminal filter, usually with a capture hood or by velocity readings across the filter face. That volume per hour is divided by the room volume. The result is compared with the air change rate specified in the HVAC design or the user requirement specification.

What is an airflow visualisation or smoke study?

A smoke study releases a visible, clean fog near critical points so the direction and behaviour of the air can be observed and video-recorded. It shows whether air sweeps away from the product and towards the returns, and highlights turbulence or dead zones. Regulators commonly expect it for aseptic areas under EU GMP Annex 1.

Which standard describes cleanroom airflow test methods?

ISO 14644-3 sets out the test methods for cleanrooms and clean zones, including airflow volume and velocity measurement and airflow direction visualisation. In pharmaceutical facilities the results are also assessed against GMP expectations such as EU GMP Annex 1, WHO guidance on HVAC systems and Schedule M, together with the site's own design specification.

Can the test be done without shutting down production?

Most measurements are taken at terminal filters, grilles and openings, so they can usually be scheduled around production with limited disruption. Smoke studies in aseptic areas are normally planned with the site QA team, because they may be required in both at-rest and operational conditions and need careful control of the fog source.
